The brothers could be going up against the Men of Letters, the group they thought they were a part of.
In the season 11 finale, Lady Toni Bevell, a London branch member of the Men of Letters managed to capture Sam. At that point though, Sam thought Dean was already dead which made him lose his will to fight.

The idea of the brothers fighting mortal enemies would be a fresh approach for the show. Fans have been used to the idea of the Winchester brothers going up against all sorts of supernatural entities.

A Resurrected Mary?
