There have been speculations that the next MCU flick will finally reveal the fifth Infinity Stone, since "Captain America: Civil War" had made a point of skipping that particular plotline. Now it looks like the man who designed the Eye of Agamotto has confirmed that the amulet holds a very special stone (via ComicBook). This would certainly make sense as to how Benedict Cumberbatch's Sorcerer Supreme could become so powerful. But does this also mean that Josh Brolin's Thanos is coming to "Doctor Strange" to retrieve it?
Although it is highly unlikely that Thanos will be the main villain in "Doctor Strange", it is possible that the Mad Titan will make an appearance to tie the film with the main MCU plotline. Also, Thanos has made an appearance every time a new Infinity Stone was introduced after the Aether in the second "Thor" film. If "Doctor Strange" will confirm that the Eye of Agamotto contains the Time Stone, it wouldn't be a surprise if the Mad Titan will suddenly appear in the movie.

The actual Infinity Stone that could be hidden in the Eye of Agamotto has not yet been confirmed. Strictly speaking, prop master Barry Gibbs did not verify that the stone is actually one of the gems Thanos is looking for. However, there are speculations that the amulet will contain the Time Stone because Benedict Cumberbatch's Sorcerer Supreme has the ability to manipulate time in "Doctor Strange".
